Product Specific Information

This 7-239 monoclonal antibody reacts with human CD169 (Siglec-1, Sialoadhesin), a type I transmembrane protein of the sialic acid binding Ig-like lectin (Siglec) family.
CD169 is expressed on macrophages and dendritic cells in peripheral blood, lymph nodes, and spleen. It binds glycoproteins and glycolipids containing N-acetylneuraminyl α2-3-galactose.
Expression can be induced on CD14+ monocytes by interferon-alpha and interferon-gamma, mediating HIV-1 infection. It also plays a role in antigen presentation to invariant NK T cells.

This antibody inhibits erythrocyte binding to CD169-expressing cells.
Reported for use in flow cytometric analysis; pre-titrated and tested on normal human peripheral blood cells.
Recommended usage: 5 µL (0.5 µg) per test, staining 10⁵–10⁸ cells in 100 µL final volume.

PE-eFluor® 610 excitation/emission: 488–561 nm / 607 nm.
Recommended filter: 610/20 band pass (equivalent to PE-Texas Red®).
Ensure instrument compatibility with this fluorochrome.

Light Sensitivity: Protect from light to prevent photo-induced oxidation.
Fixation: Samples may be stored up to 3 days at 4°C in the dark using:

• IC Fixation Buffer (Product #00-8222)
• 1-step Fix/Lyse Solution (Product #00-5333)

Target Information

Siglecs are membrane-bound lectins of the immunoglobulin superfamily that bind sialic acid and mediate cell-cell interactions.
Family members include sialoadhesin, CD22, and CD33.
Sialoadhesin is a macrophage-specific adhesion molecule expressed on activated macrophages in chronic inflammation and tumors.
