Product Specific Information

Description:
The CEW33D monoclonal antibody reacts with human CD183 (CXCR3), a G protein-coupled chemokine receptor that binds CXCL9 (MIG), CXCL10 (IP-10), and CXCL11 (I-TAC). CD183 is associated with type 1 immunity and is upregulated in Th1, CD8 effector, NK, and NKT cells. It mediates infiltration of inflamed tissues in immune responses and inflammatory diseases. Also expressed on some B cells and plasmacytoid dendritic cells.

Applications Reported:
Validated for flow cytometric analysis.

Applications Tested:
Pre-titrated and tested on normal human peripheral blood cells. Use 5 µL (0.5 µg) per test (100 µL final volume). Cell number: 10⁵–10⁸ cells/test.

Fluorophore Information:
PE-eFluor® 610 is excited by 488–561 nm lasers and emits at 607 nm. Recommended filter: 610/20 band pass (equivalent to PE-Texas Red®). Protect from light due to photo-induced oxidation sensitivity.

Fixation:
Samples can be stored in:

• IC Fixation Buffer (Product #00-8222)
• 1-step Fix/Lyse Solution (Product #00-5333)
  Store up to 3 days at 4°C in the dark with minimal impact on brightness and FRET efficiency.

Target Information

This gene encodes a G protein-coupled receptor selective for chemokines IP10, Mig, and I-TAC, which belong to the CXC subfamily. Binding of these chemokines induces leukocyte trafficking, integrin activation, and chemotaxis. Expression is high in effector/memory T cells and in inflamed tissues. It is a potential therapeutic target for inflammatory diseases. Multiple transcript variants exist.
