Product Specific Information

The monoclonal antibody CXCR3-173 recognizes mouse CD183 (CXCR3), a seven-transmembrane G-protein-coupled chemokine receptor binding to CXCL9 (MIG), CXCL10 (IP-10), and CXCL11 (ITAC). CD183 is involved in CD4 T cell responses to grafts, and CXCR3 knockout mice show compromised allograft rejection. Expression is found on NK cells, subsets of T lymphocytes and Tregs, and is preferentially expressed on Th1-polarized cells.

Fluorochrome characteristics:
PE-eFluor 610 can be excited with 488–561 nm lasers and emits at 607 nm. Recommended detection filter: 610/20 band pass (PE-Texas Red equivalent). Ensure instrument compatibility.

Light sensitivity: This tandem dye is sensitive to photo-induced oxidation. Protect vial and stained samples from light.

Fixation: Samples can be stored in IC Fixation Buffer (Product #00-8222-49) or 1-step Fix/Lyse Solution (Product #00-5333-57) for up to 3 days at 4°C in the dark with minimal impact on brightness and FRET efficiency.

Target Information

This gene encodes a G protein-coupled receptor selective for the chemokines IP10, Mig, and I-TAC. These belong to the CXC chemokine subfamily, characterized by a single amino acid separating the first two cysteine residues. Chemokine binding induces cellular responses involved in leukocyte trafficking, integrin activation, cytoskeletal rearrangement, and chemotaxis. CXCR3 signaling involves Gi-type G proteins and is rapidly desensitized via receptor internalization. Expression is prominent in effector/memory T cells and inflamed tissues, suggesting a role in inflammatory cell recruitment. CXCR3 is a potential target for small molecule antagonists in inflammatory disease research.
