Product Specific Information

The A2MR-a2 monoclonal antibody reacts with human CD91, also known as LRP-1 and alpha-2 macroglobulin receptor. CD91 is a 600 kDa type I transmembrane protein composed of alpha and beta chains. The alpha chain binds multiple ligands, while the beta chain contains two tyrosine phosphorylation motifs involved in signal transduction. CD91 mediates ligand endocytosis and lysosomal trafficking and is expressed in monocytes, dendritic cells, hepatocytes, fibroblasts, adipocytes, neurons, and syncytiotrophoblasts. It plays a role in antigen cross-presentation via MHC class I, cell migration, and intracellular signaling.

Applications Reported

This antibody is reported for use in flow cytometric analysis.

Applications Tested

Pre-titrated and tested by flow cytometric analysis of normal human peripheral blood cells.
Use 5 µL (0.05 µg) per test in a final volume of 100 µL.
Cell number should be empirically determined (10⁵–10⁸ cells/test).

Important Notes

• NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability.
• CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) is required for optimal staining.
  • Add 5 µL per sample before combining with cells.
  • For single-color controls, use 5 µL per 100 µL of cell sample (10³–10⁸ cells).

Target Information

CD91 (LRP-1) is an endocytic receptor involved in intracellular signaling, lipid homeostasis, and clearance of apoptotic cells. It is essential for A2M-mediated clearance of amyloid precursor protein and beta-amyloid, which are associated with Alzheimer’s disease. Expression decreases with age and is reduced in Alzheimer’s brain tissue.
