Target Information

X-linked ectodysplasin-A2 receptor (XEDAR) is a member of the tumor necrosis factor receptor family, highly expressed during embryonic development, and binds to ectodysplasin-A2 (EDA-A2). Two major isoforms, XEDAR-s and XEDAR-L, differ by a 21-amino-acid region at the juxtamembrane region of the cytoplasmic domain. Both isoforms lack a death domain and signal mainly through TRAF3 and TRAF6 to activate NF-κB and JNK pathways. Cells expressing XEDAR and treated with EDA-A2 form a complex containing FADD, caspase-8, and caspase-10, leading to activation of caspase-8 and caspase-3 and resulting in apoptosis. EDA-A2-induced apoptosis depends on caspase-9 activation, as inhibitors of caspase-8 block apoptosis following EDA-A2 treatment.
