Target Information
Cytochrome P450 2D6 (CYP2D6)는 cytochrome P450 초과 가족에 속하는 효소로, 약물 대사 및 콜레스테롤, 스테로이드, 지질 합성에 관여하는 monooxygenase입니다. 이 단백질은 소포체에 위치하며, 일반적으로 처방되는 약물의 약 20%를 대사합니다. 주요 기질에는 debrisoquine, sparteine, propafenone, amitryptiline 등이 포함됩니다.
CYP2D6 유전자는 다형성이 높으며, 특정 대립유전자는 약물 대사 능력이 감소된 ‘poor metabolizer’ 표현형을 유발할 수 있습니다. 해당 유전자는 22q13.1 염색체 근처의 두 개의 P450 유사유전자와 인접해 있습니다. 또한, 다양한 isoform을 암호화하는 대체 스플라이싱 전사체 변이체가 보고되었습니다.
