Product Specific Information

• Reported for intracellular staining followed by flow cytometric analysis of normal human peripheral blood cells.
• Pre-diluted and tested for use at 5 µL (0.5 µg) per test, defined as the amount that stains a cell sample in 100 µL.
• Recommended cell number: 10⁵–10⁸ cells/test (empirically determined).
• Fluorochrome characteristics:
  • Excitation: 532 nm (Green laser)
  • Emission: 561 nm
  • Not compatible with Yellow-Green laser (561 nm)
  • Recommended filter: 560/14 band pass

Target Information

Granzyme B is a serine protease found in cytotoxic T cells and NK cells, playing a crucial role in apoptosis induction of target cells. It acts via:

• Cleaving caspase-3 to initiate the caspase cascade.
• Activating mitochondrial apoptosis through Bid protein cleavage.
• Binding mannose 6-phosphate receptor as a death receptor during cytotoxic T cell-induced apoptosis.
  Granzymes (A–G in mouse, fewer in human) are stored in specialized lytic granules of cytotoxic lymphocytes.
