Target Information
GAPDH (Glyceraldehyde-3-phosphate dehydrogenase)는 해당과정(glycolysis)에 관여하는 촉매 효소로, 37 kDa의 동일한 서브유닛 4개로 구성된 테트라머 형태로 존재합니다. NADPH 존재하에 1,3-bisphosphoglycerate를 glyceraldehyde 3-phosphate로 환원시키는 반응을 촉매합니다.
GAPDH는 대부분의 조직에서 높은 수준으로 발현되며, glycolysis 외에도 DNA 복제 및 수선, RNA 수출, 막 융합, 미세소관 결합 등 다양한 세포 기능에 관여합니다. 또한 신경퇴행성 질환, 세포사멸(apoptosis), 암 등에서 유전자 발현 및 세포 표현형 조절에 중요한 역할을 하는 것으로 보고되었습니다.
GAPDH는 Alzheimer’s disease와 관련된 amyloid precursor protein, Huntington’s disease의 Huntingtin 단백질 등 여러 단백질과 결합하는 것으로 알려져 있습니다. 높은 발현 수준으로 인해 Western blot에서 단백질 로딩 컨트롤로 널리 사용됩니다.
