Product Specific Information
For reconstitution, add 100 µL of distilled water to achieve a final antibody concentration of approximately 1 mg/mL.
For conjugation experiments, perform an additional desalting step using Zeba Spin Desalting Columns (7K MWCO, 0.5 mL, Product #89882).
Target Information
The tumor suppressor protein p53 is a sequence-specific transcription factor activated by cellular stress. It mediates cell cycle arrest or apoptosis in response to DNA damage or nucleotide starvation.
p53 activation leads to transcription of genes such as p21waf1, contributing to G1 or G2/M arrest. Structurally, p53 includes:
- N-terminal transactivation domain
- Central DNA-binding domain
- Oligomerisation domain
- C-terminal regulatory domain
Phosphorylation at Ser15 is crucial for activation and stabilization, while phosphorylation at Ser392 correlates with tumor formation.
p53 plays a vital role in preventing proliferation of damaged cells, acting as a natural anticancer agent. It is also associated with aging, oxidative stress, and cognitive functions such as long-term potentiation (LTP).
Mutations in TP53 can lead to loss of tumor suppressor function, contributing to malignancies and hereditary cancer syndromes like Li-Fraumeni syndrome.
