Description:
The OX39 monoclonal antibody reacts with mouse CD25, the 55 kDa interleukin-2 receptor alpha (IL-2Rα). CD25 is expressed by early progenitors of T and B lineages and activated mature T and B lymphocytes. CD25 binds IL-2 with low affinity but forms a high-affinity receptor when associated with CD122 (IL-2Rβ) and CD132 (common γ chain). CD25 plays a role in lymphocyte differentiation and activation/proliferation.

The OX39 clone has been shown to deplete CD25 cells in vivo.

Applications Reported:
This antibody is suitable for flow cytometric analysis.

Applications Tested:
Tested by flow cytometric analysis of rat splenocytes. Recommended ≤ 0.06 µg/test (100 µL final volume). Optimize antibody titration for best performance.

Fluorochrome Information:
PerCP-eFluor® 710 emits at 710 nm and is excited at 488 nm (blue laser). Recommended filter: 710/50 bandpass (alternative: 695/40 bandpass).

Fixation:
Samples may be stored in IC Fixation Buffer (Product #00-8222) or 1-step Fix/Lyse Solution (Product #00-5333) for up to 3 days at 4°C in the dark with minimal impact on brightness and FRET efficiency.

Target Information

CD25 (IL2RA):
CD25 encodes the IL-2 receptor alpha chain, part of a heterotrimeric receptor complex (IL2RA, IL2RB, IL2RG) forming the high-affinity IL-2 receptor. IL2RA homodimers form low-affinity receptors. IL2 is mainly produced by mature T cells and regulates proliferation, differentiation, and apoptosis of lymphocytes. Mutations in IL2RA are associated with interleukin-2 receptor alpha deficiency and immune dysregulation.
