Product Specific Information

The 1C10 monoclonal antibody reacts with mouse CD40, a 45–50 kDa type I transmembrane glycoprotein. CD40 is a TNFR family member expressed on mouse B lymphocytes, follicular dendritic cells, thymic epithelium, and some peripheral T cells.
CD40 regulates B cell development and maturation by inducing Ig isotype switching and preventing apoptosis in combination with IL-4. Interaction with CD154 (gp39) is critical for T-B cell crosstalk and immune regulation.

The 1C10 antibody exhibits agonistic activity in vitro and in vivo.

Fluorophore Information:
PerCP-eFluor® 710 emits at 710 nm, excited by blue laser (488 nm).
Recommended filter: 710/50 bandpass (695/40 acceptable alternative).
Ensure instrument compatibility.

Fixation:
Samples may be stored in IC Fixation Buffer (Product #00-8222) or 1-step Fix/Lyse Solution (Product #00-5333) for up to 3 days at 4°C in the dark.

Target Information

CD40 antigen is a single-chain glycoprotein, part of the TNF/NGF receptor superfamily. It is expressed on B cells (except plasma cells), epithelial cells, carcinomas, and lymphoid dendritic cells.
CD40 is essential for immune responses including T cell-dependent Ig class switching, memory B cell development, and germinal center formation.
Regulated by transcription factor AKNA and interacts with TNFR2 for signal transduction.
Associated with amyloid-beta-induced microglial activation and Alzheimer’s disease pathogenesis.
Two alternatively spliced transcript variants have been reported.
Diseases linked to CD40 dysfunction include Type 3 Hyper-IgM immunodeficiency and CD40 ligand deficiency.
