Product Specific Information
Description:
The 16-10A1 monoclonal antibody reacts with mouse CD80 (B7-1), a 55 kDa member of the Ig superfamily. CD80 is expressed by macrophages, dendritic cells, activated B cells, and activated T cells. It binds to CD28 and CD152 (CTLA-4) on T cells, playing a key role in T-B cell communication and activation.
Applications Reported:
Reported for use in flow cytometric analysis.
Applications Tested:
Tested by flow cytometric analysis of mouse splenocytes. Recommended ≤0.125 µg/test (100 µL final volume). Optimal titration is advised.
Fluorochrome Information:
Brilliant Violet™ 605 (BV605) emits at 605 nm and is compatible with violet (405 nm) laser-equipped cytometers. Protect from light due to photo-induced oxidation sensitivity.
Staining Recommendations:
When using multiple polymer dye-conjugated antibodies (e.g., Super Bright, Brilliant Violet™, Brilliant Ultra Violet™), use Super Bright Complete Staining Buffer (#SB-4401-42) or Brilliant Stain Buffer™ (#00-4409-75) to minimize non-specific interactions.
Fixation:
Samples can be stored in IC Fixation Buffer (#00-8222-49) or 1-step Fix/Lyse Solution (#00-5333-54) up to 3 days at 4°C in the dark. BV605 is not compatible with methanol-based fixation.

Target Information
CD80 (B7-1) and CD86 (B7-2) are ligands for T cell costimulatory molecule CD28 and inhibitory receptor CTLA-4 (CD152). Both are expressed on antigen-presenting cells and are essential for T cell activation. CD80 is induced on activated B cells, EBV-transformed B cell lines, Burkitt lymphoma cells, T cells, and monocytes. It plays a regulatory role in T lymphocyte activation via CD28 and CTLA-4 binding.
Diseases associated with CD80 dysfunction include gallbladder squamous cell carcinoma and myocarditis.
