This antibody specifically detects recombinant proteins containing the seven amino acid 6xHis-Gly epitope, commonly found in expression vectors encoding an N-terminal 6xHis tag.
The HisG antibody recognizes the sequence -His-His-His-His-His-His-Gly and is suitable for detecting fusion proteins from bacterial, insect, and mammalian cells.

Anti-HisG-HRP Antibody was prepared by crosslinking the Anti-HisG Antibody with horseradish peroxidase using glutaraldehyde. It has been validated in immunoblotting and ELISA procedures using purified Positope™ control protein (5 ng).
Positope™ is a 53 kDa recombinant protein containing multiple epitope tags, including His(C-term), HisG, c-Myc, and V5. Low background was observed using chemiluminescent or alkaline phosphatase detection reagents.

For western blot:

• Dilute in PBS containing 0.05% Tween-20 and 5% nonfat dry milk (PBSTM).
• No cross-reactivity observed in bacterial lysates; minor cross-reactivity in mammalian lysates upon overexposure.
• If using alkaline phosphatase-conjugated secondary antibody, use TBS instead of PBS (phosphate inhibits alkaline phosphatase).
• Wash blots or wells before adding HRP-conjugated antibody.
• Avoid azide (inhibits HRP activity).

This product contains sufficient material for approximately 25 western blots (assuming 10 mL buffer per blot).

Target Information

Epitope tags enable localization and characterization of gene products, protein complexes, and low-abundance proteins when specific antibodies are unavailable.
Expression of recombinant proteins with histidine residues in E. coli facilitates purification via Ni²⁺-NTA affinity chromatography.
HisG antibodies detect recombinant proteins containing the polyhistidine sequence His-His-His-His-His-His-Gly (6xHis-Gly epitope) and are applicable to bacterial, insect, and mammalian expression systems.
