Product Specific Information

Description:
The eBio124-1D1 monoclonal antibody reacts with human CD7 (gp40, Leu9), a 40 kDa receptor belonging to the immunoglobulin gene superfamily. The N-terminal sequence (aa1-107) is homologous to the Ig kappa light chain, while the C-terminal region is proline-rich and forms a stalk-like structure. CD7 is expressed on most immature and mature T lymphocytes, T cell leukemias, NK cells, a small subset of B cells, and malignant B cells.

Cross-linking of surface CD7 enhances T cell and NK cell activity, influencing calcium flux, adhesion molecule expression, cytokine secretion, and proliferation. CD7 associates with phosphoinositol 3'-kinase, and its ligation induces D-3 phosphoinositide production and tyrosine phosphorylation.

A clonogenic subpopulation of CD34(+) CD38(-) cord blood cells expressing high levels of CD7 has lymphopoietic potential, generating B cells, NK cells, and dendritic cells. CD7(+) phenotype distinguishes primitive lymphoid progenitors from pluripotent stem cells. CD7 also cooperates with CD28 during Treg function, as deficiency in both reduces Treg numbers and suppressive activity.

Each product includes one vial of NovaFluor conjugate and one vial of CellBlox Plus Blocking Buffer.

Applications Reported:
Validated for flow cytometric analysis.

Applications Tested:
Pre-titrated and tested on normal human peripheral blood cells. Use 5 µL (0.8 µg) per test in 100 µL final volume. Cell number range: 10⁵–10⁸ cells/test.

Important Notes:

  • NovaFluor dyes are incompatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
  • Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability assays.
  • Use CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) for optimal staining. Add 5 µL per sample before combining with cells.
  • For single-color controls: 5 µL of blocking buffer per 100 µL of cell sample (10³–10⁸ cells).
  • NovaFluor conjugates utilize Phiton™ technology for engineered fluorescence with minimized spillover and spread.

CD7 (gp40, Leu9) is a 40 kDa member of the immunoglobulin gene superfamily. It is expressed on most immature and mature T lymphocytes, T cell leukemias, NK cells, and subsets of B cells. CD7 cross-linking modulates T and NK cell function through calcium signaling and cytokine regulation. It is a key marker in leukemia diagnostics.
