Product Specific Information

The monoclonal antibody phpu13 recognizes human and mouse PU.1, a member of the ETS family of transcription factors. PU.1 (42 kDa) was originally identified as Spi-1, a proto-oncogene involved in Friend virus-induced murine erythroleukemia. Its expression plays a crucial role in lymphocyte and myeloid development. Knock-out mice lacking PU.1 are neonatal lethal and deficient in myeloid, T, and B cells, with altered erythroid development.

Protein expression levels are essential for proper development, showing higher levels in myeloid cells compared to lymphocytes. PU.1 regulates transcription of several proteins including CD11b, CD18, Gata2, IRF4, CD20, CD10, and CD79.

Disruption of PU.1 expression has been associated with various leukemias, including:

  • B-chronic lymphocytic leukemia
  • Mantle cell lymphoma
  • Follicular lymphoma
  • Marginal zone lymphoma
  • Burkitt lymphoma
  • Diffuse large cell lymphoma
  • Diffuse large B-cell lymphoma
  • T-cell rich B-cell lymphoma
  • Nodular lymphocyte predominant Hodgkin lymphoma

Applications Reported: Suitable for immunohistochemical staining of formalin-fixed paraffin-embedded tissue sections and immunocytochemistry.

Applications Tested: Verified by IHC on formalin-fixed paraffin-embedded tissue using IHC Antigen Retrieval Solution - Low pH (10X) (Product #00-4955-58). Recommended concentration ≤10 µg/mL, with titration for optimal assay performance.

Target Information

This gene encodes an ETS-domain transcription factor that activates gene expression during myeloid and B-lymphoid cell development. The nuclear protein binds to a purine-rich PU-box near promoters of target genes and regulates their expression with other transcription factors and cofactors. It also influences alternative splicing of target genes. Multiple transcript variants encoding different isoforms have been identified.
