Product Specific Information
The 53-6.7 monoclonal antibody reacts with the mouse CD8a molecule, a 32–34 kDa cell surface receptor expressed as CD8αβ or CD8αα dimers.
- CD8αβ: Found on thymocytes and mature αβ TCR T cells
- CD8αα: Found on γδ TCR T cells, intestinal intraepithelial lymphocytes, and dendritic cells
CD8 binds to MHC class I and associates with p56lck, playing a key role in T-cell development and activation.
Applications Reported: Immunohistochemistry (Frozen) and Immunocytochemistry
Applications Tested: IHC-F on mouse spleen ≤ 5 µg/mL
Filter Recommendation: Use filters capturing 615 nm emission (e.g., Excitation 560/55, 585LP, Emission 645/75). Alexa Fluor 594 filters are acceptable.

CD8 (Cluster of Differentiation 8) is a type I transmembrane glycoprotein of the immunoglobulin family that functions as a co-receptor for MHC class I molecules.
- Expressed on T cells as disulfide-linked heterodimers (CD8αβ) or homodimers (CD8αα)
- Involved in T cell differentiation, activation, and signal transduction
- CD8 interaction with MHC-I triggers Lck recruitment, leading to cytokine production and cytotoxic T lymphocyte (CTL) activation
- CTLs are critical for pathogen and tumor clearance
- Differentiation of naive CD8+ T cells into CTLs is enhanced by IL-2, IL-12, and TGF-β1
