Product Specific Information

Description:
The IP26 monoclonal antibody reacts with the alpha beta chain of human TCR. The alpha beta TCR is expressed by the majority of peripheral T cells.

Applications Reported:
This IP26 antibody has been reported for use in flow cytometric analysis.

Applications Tested:
This IP26 antibody has been pre-titrated and tested by flow cytometric analysis of normal human peripheral blood cells. Recommended use at 5 µL (0.5 µg) per test (100 µL final volume). Cell number should be determined empirically (10⁵–10⁸ cells/test).

Fluorochrome Information:
PerCP-eFluor® 710 emits at 710 nm and is excited with the blue laser (488 nm). It can be used in place of PerCP-Cyanine5.5.
Recommended filter: 710/50 bandpass (695/40 acceptable alternative). Ensure instrument compatibility.

Fixation:
Samples can be stored in IC Fixation Buffer (Product #00-8222-49) or 1-step Fix/Lyse Solution (Product #00-5333-54) for up to 3 days at 4°C in the dark with minimal impact on brightness and FRET efficiency.

Excitation/Emission:

• Excitation: 488 nm

0.2 µm post-manufacturing filtered.

Target Information

The T cell antigen receptor (TCR) consists of a ligand-specific alpha/beta heterodimer associated with invariant CD3 subunits, essential for surface expression and signal transduction.
TCR engagement triggers T cell activation, differentiation, and cytokine production.
Most T cells express TCR alpha/beta heterodimers, while gamma/delta T cells are mainly found in epithelial tissues.
Defects in TCR expression or recognition can contribute to autoimmune diseases and tumor-related immune responses.
Superantigen binding to TCRs is linked to toxic shock syndrome.
Autoantibodies against TCR segments have been observed in autoimmune diseases such as RA and SLE.
