Description:
OX1 monoclonal antibody recognizes a non-polymorphic determinant of rat CD45 (leukocyte common antigen). CD45 is expressed on all hematopoietic cells except erythrocytes and platelets. It is a type I transmembrane protein tyrosine phosphatase with multiple isoforms generated by alternative splicing.

Applications Reported:
Reported for use in flow cytometric analysis.

Applications Tested:
Tested by flow cytometric analysis of rat splenocytes. Recommended ≤ 1 µg/test (100 µL final volume). Cell number may range from 10⁵–10⁸ cells/test. Antibody titration is recommended for optimal performance.

Fluorophore Information:
APC-eFluor 780 emits at 780 nm, excited by Red laser (633 nm). Ensure instrument compatibility.

Light Sensitivity:
Sensitive to photo-induced oxidation. Protect vial and stained samples from light.

Fixation:
Samples can be stored in IC Fixation Buffer (Product #00-8222) or 1-step Fix/Lyse Solution (Product #00-5333) for up to 3 days at 4°C in the dark with minimal impact on brightness and FRET efficiency. Clone-specific performance should be determined empirically.

Target Information

CD45 (LCA, leukocyte common antigen) is a receptor-type protein tyrosine phosphatase expressed in all nucleated hematopoietic cells (~10% of lymphocyte surface proteins). Absent in non-hematopoietic cells and tissues. CD45 regulates Src-family kinases, antigen receptor signaling, and cytokine receptor pathways. Isoforms differ by splicing and cell type. CD45 also modulates cell survival, integrin signaling, and apoptosis.
