Product Specific Information

Description:
The HI100 monoclonal antibody reacts with human CD45RA, a 220 kDa molecule expressed by subsets of CD4+ and CD8+ peripheral T lymphocytes, as well as B cells. CD45RA+ T cells are primarily naive/virgin, making the HI100 mAb a useful marker for distinguishing T cell subsets.

Applications Reported:
This antibody is reported for use in flow cytometric analysis.

Applications Tested:
Pre-titrated and validated for flow cytometry using normal human peripheral blood cells. Recommended use: 5 µL (0.25 µg) per test (100 µL final volume). Cell number range: 10⁵–10⁸ cells/test.

Fluorophore Information:
APC-eFluor 780 emits at 780 nm and is excited by the red laser (633 nm). Ensure instrument compatibility.

Light Sensitivity:
Sensitive to photo-induced oxidation. Protect from light during storage and staining.

Fixation:
Samples can be stored in:

• IC Fixation Buffer (Product #00-8222)
• 1-step Fix/Lyse Solution (Product #00-5333)
  for up to 3 days at 4°C in the dark with minimal impact on brightness and FRET efficiency.

Target Information

CD45RA is an isoform of the CD45 complex with restricted expression among lymphoid cell subtypes. CD45 (LCA, leukocyte common antigen) is a receptor-type protein tyrosine phosphatase expressed on all nucleated hematopoietic cells (~10% of lymphocyte surface proteins). It regulates Src-family kinases and plays roles in lymphocyte development, antigen signaling, and integrin-mediated pathways.
CD45 isoforms differ by alternative splicing, affecting extracellular domains while sharing identical transmembrane and cytoplasmic regions.
CD45 dysfunction is associated with severe combined immunodeficiency and Hepatitis C.
