TFAM interacts with autophagy proteins such as LC3 via its LC3 interacting region (LIR) motif, aiding in the removal of leaked mtDNA and controlling inflammation.
TFAM levels influence mitochondrial function across tissues — high levels can repress mtDNA expression in skeletal muscle, while in the heart, balanced TFAM-to-mtDNA ratios preserve oxidative phosphorylation (OXPHOS) capacity.
Disruptions in TFAM function are linked to mitochondrial dysfunction and diseases related to mtDNA stress, including neurodegeneration.
