Target Information
Monomethyl auristatin F (MMAF) is a synthetic antineoplastic agent used in the approved drug belantamab mafodotin for multiple myeloma and in experimental antibody-drug conjugates such as vorsetuzumab mafodotin and SGN-CD19A.
In International Nonproprietary Names for MMAF-antibody conjugates, "mafodotin" refers to MMAF plus its attachment structure to the antibody.
MMAF acts as an antimitotic agent by inhibiting tubulin polymerization, thereby blocking cell division. When linked to antibodies targeting cancer cell structures, MMAF accumulates selectively in those cells.
Chemically, MMAF is desmethyl-auristatin F, meaning its N-terminal amino group has only one methyl substituent instead of two as in auristatin F.
