Product Specific Information

The 4H11 monoclonal antibody reacts with human CD34 (mucosialin), a transmembrane protein expressed on hematopoietic progenitors, mesenchymal stem cells, tumor cell lines, and vascular endothelia. CD34 epitopes are classified into three types (I, II, III); 4H11 belongs to class III, reacting with a protein epitope.

Applications Reported: Suitable for flow cytometric analysis.
Applications Tested: Pre-titrated and tested with normal human peripheral blood cells at 5 µL (0.25 µg) per test.
A test is defined as the amount of antibody staining a cell sample in 100 µL volume (10⁵–10⁸ cells/test).

Fluorochrome Information:
Super Bright 600 is a tandem dye excited by the violet laser (405 nm) and emits at 600 nm. Recommended detection filter: 610/20 bandpass. Ensure instrument compatibility.

Staining Buffer Recommendation:
When using multiple Super Bright-conjugated antibodies, use Super Bright Complete Staining Buffer (Product # SB-4401) to minimize non-specific polymer interactions.

Light Sensitivity:
Sensitive to photo-induced oxidation. Protect from light.

Fixation:
Samples can be stored in IC Fixation Buffer (Product # 00-8222) or 1-step Fix/Lyse Solution (Product # 00-5333) for up to 3 days at 2–8°C in the dark with minimal impact on brightness.

Target Information

CD34 is a highly glycosylated surface protein (111–115 kDa) present on stem cell populations. It serves as a stem cell marker and may regulate adhesion, differentiation, and proliferation. CD34 expression reflects specific hematopoietic development stages and may mediate stem cell attachment to the bone marrow matrix or stromal cells. It may also present carbohydrate ligands to selectins and participate in signal transduction via phosphorylation by protein kinase C.
Diseases associated with CD34 dysfunction include dermatofibrosarcoma and neurofibroma.
