Product Specific Information

Description:
The WM-53 monoclonal antibody reacts with human CD33 (GP67, P67), a 67 kDa type I transmembrane glycoprotein of the Siglec family. It is expressed on monocytes, activated T cells, granulocytes, myeloid progenitors, and mast cells.

Applications Reported:
This WM-53 antibody is suitable for flow cytometric analysis.

Applications Tested:
Pre-diluted and validated for flow cytometry using normal human peripheral blood cells. Recommended usage: 5 µL (0.25 µg) per test (100 µL total volume, 10⁵–10⁸ cells/test).

Fluorochrome Information:
Super Bright 600 is a tandem dye excited by a 405 nm violet laser and emits at 600 nm. Use a 610/20 bandpass filter. Ensure instrument compatibility.

Staining Recommendation:
When using multiple Super Bright dyes, use Super Bright Complete Staining Buffer (Product # SB-4401) to minimize non-specific interactions.

Fixation:
Samples can be fixed using IC Fixation Buffer (Product # 00-8222-49) or 1-step Fix/Lyse Solution (Product # 00-5333-57) for up to 3 days at 4°C in the dark with minimal signal loss.

Light Sensitivity:
Protect from light to prevent photo-induced oxidation.

Target Information

CD33 (Siglec-3):
CD33 is a transmembrane protein in the sialic acid-binding immunoglobulin-like lectin (Siglec) family containing ITIM motifs that recruit SHP-1 and SHP-2 phosphatases. It is expressed on myelomonocytic lineage cells, including granulocyte and macrophage precursors, and serves as a therapeutic target for acute myeloid leukemia. CD33 is also a useful marker for distinguishing myelogenous leukemia from lymphoid or erythroid leukemias.
