Target Information
Peroxiredoxin (Prx) is a family of peroxidases associated with cell proliferation, differentiation, and apoptosis. Mammalian Prx proteins (approximately 50 isoforms) are classified by amino acid sequence homology and contain active site cysteine residues with thiol-specific antioxidant activity.
Mammalian Prxs consist of six members grouped into typical 2-Cys, atypical 2-Cys, and 1-Cys subfamilies. Except for Prx VI (1-Cys type), the other five 2-Cys Prx isotypes exhibit thioredoxin-dependent peroxidase (TPx) activity using thioredoxin, thioredoxin reductase, and NADPH as a reducing system.
Mammalian Prxs are 20–30 kDa in size and differ in subcellular localization: Prx I, II, and VI are cytosolic; Prx III is mitochondrial; Prx IV is ER-associated and secreted; Prx V is found in peroxisomes, mitochondria, and cytosol.
