Target Information
CD20 is a non-glycosylated surface phosphoprotein with a molecular weight of 33–37 kDa depending on phosphorylation. It is expressed on mature and most malignant B cells, as well as a subset of T lymphocytes and follicular dendritic cells. CD20 expression coincides with surface IgM and regulates calcium conductance, cell cycle progression, and B-cell proliferation.
CD20 is associated with lipid rafts, with its association influenced by extracellular signals and BCR aggregation. Upon receptor ligation, CD20 colocalizes with BCR before dissociation, remaining on the cell surface. CD20 is a key target for antibody-mediated B-cell depletion, as it is highly expressed on B-cell malignancies and remains on the plasma membrane after treatment. Dysfunction of CD20 is associated with MS4A1-related common variable immune deficiency.
