Product Specific Information
The XMG1.2 antibody reacts with mouse interferon (IFN) gamma and acts as a neutralizing antibody. IFN gamma is a 20 kDa cytokine produced by activated T, B, and NK cells, exhibiting antiviral and antiparasitic activity. It synergizes with TNF alpha to inhibit cell proliferation and modulates immune responses across various cell types expressing IFN gamma receptors. Glycosylation does not affect its biological activity.
Applications Reported: Intracellular staining followed by flow cytometric analysis.
Applications Tested: Verified using mouse splenocytes and the Intracellular Fixation & Permeabilization Buffer Set (Product # 88-8824-00). Recommended usage ≤1.0 µg/test (100 µL final volume).
Brilliant Violet™ 650 (BV650) emits at 649 nm and is suitable for cytometers equipped with a 405 nm violet laser. Use Super Bright Complete Staining Buffer (Product # SB-4401-42) or Brilliant Stain Buffer™ (Product # 00-4409-75) to minimize non-specific polymer interactions.
Light Sensitivity: Protect from light to prevent photo-induced oxidation.
Fixation: Compatible with IC Fixation Buffer (Product # 00-8222-49) or 1-step Fix/Lyse Solution (Product # 00-5333-54). Not compatible with methanol-based fixation.

Target Information
IFN gamma (Interferon gamma, Type II interferon) is a macrophage activation and immune interferon produced mainly by T-lymphocytes and NK cells. It is a dimeric glycoprotein (~45 kDa) with antiviral, antiproliferative, and immunomodulatory functions. Human IFN gamma does not cross-react with mouse IFN gamma. It activates macrophages, induces MHC expression, and enhances B-cell immunoglobulin secretion via the JAK-STAT pathway. Mutations in the IFN gamma gene are associated with aplastic anemia.
