Product Specific Information
The monoclonal antibody 3E4 reacts with human and mouse interferon regulatory factor 4 (IRF4). IRF4 is a 52 kDa transcription factor involved in B cell, T cell, and macrophage function. It is highly expressed in mature plasma cells and regulates differentiation and gene transcription such as Prdm1 (Blimp1). In T cells, IRF4 influences Treg, Th2, Th9, and Th17 development and function, inducing IL-10 expression. IRF4 also regulates macrophage polarization and is upregulated in various hematologic malignancies.
Applications Reported: Intracellular staining followed by flow cytometric analysis.
Applications Tested: Tested with normal human peripheral blood cells using Foxp3/Transcription Factor Staining Buffer Set (Product #00-5523-00). Recommended use ≤1.0 µg/test (100 µL final volume). Cell number range: 10⁵–10⁸ cells/test. Optimize antibody titration for best results.
Fluorochrome Information:
Brilliant Violet™ 650 (BV650) emits at 649 nm, suitable for cytometers with a violet (405 nm) laser. Ensure instrument compatibility.
Buffer Recommendations:
Use Super Bright Complete Staining Buffer (Product #SB-4401-42) or Brilliant Stain Buffer™ (Product #00-4409-75) to reduce polymer dye interactions in multi-color panels.
Handling Notes:
- Light sensitivity: Protect vial and samples from light.
- Fixation: Samples can be stored up to 3 days at 4°C in dark using IC Fixation Buffer (Product #00-8222-49) or 1-step Fix/Lyse Solution (Product #00-5333-54).
- Methanol-based fixation is incompatible with BV650.

Target Information
The IRF (interferon regulatory factor) family of transcription factors possess a unique tryptophan pentad repeat DNA-binding domain. IRFs regulate interferon responses and interferon-inducible genes. IRF4 is lymphocyte-specific and negatively regulates TLR signaling, crucial for innate and adaptive immune activation. A chromosomal translocation t(6;14)(p25;q32) involving IRF4 and IgH may cause multiple myeloma. Alternative splicing variants exist.
