Description:
The MIH18 monoclonal antibody reacts with human B7-DC, also known as PD-L2. B7-DC is a member of the B7 family with a predicted molecular weight of approximately 25 kDa and belongs to the immunoglobulin superfamily. It is primarily expressed by dendritic cells and monocytes/macrophages. Unlike other B7 family members, B7-DC does not bind CD28/CTLA-4 but acts as a ligand for PD-1.

Interactions between PD-1 and B7-DC/PD-L2 are involved in either costimulation or suppression of T cell proliferation depending on cellular activation. MIH18 is a blocking antibody useful for studying B7-DC/PD-L2 function and expression patterns in human immune cells.

Target Information

Programmed death-ligand 2 (PD-L2, B7-DC) is part of the B7 ligand family within the immunoglobulin superfamily. Alongside PD-L1, it acts as a ligand for PD-1. PD-L2 is expressed mainly in dendritic cells but can be induced in various immune and non-immune cells depending on the microenvironment.

PD-L2 expression is upregulated by Th2 cytokine IL-4 and, to a lesser extent, by Th1 cytokines TNF-α and IFN-γ. Although expressed at lower levels than PD-L1, PD-L2 has 2–6× higher affinity for PD-1. PD-L2 and PD-L1 serve as inhibitory immune checkpoint molecules that regulate T cell activation and cytokine secretion (e.g., IFN-γ, IL-10).

Interestingly, PD-L2 can also stimulate T cell proliferation and cytokine production even in PD-1-deficient T cells, suggesting additional receptor interactions. PD-L2 also binds to RGMb (repulsive guidance molecule b), a receptor involved in BMP signaling and respiratory tolerance regulation in the lung.
