Metadherin (MTDH, LYRIC)
Metadherin, also known as MTDH or LYRIC (LYsine-RIch CEACAM1 co-isolated), is a protein localized with tight junction proteins ZO-1 and occludin in polarized epithelial cells. It is recruited during tight junction maturation rather than serving as a structural component.
Metadherin is overexpressed in breast cancer tissues and xenografts, with lower expression in normal breast tissue. It binds to lung vasculature, one of the main sites of breast cancer metastasis. Blocking its lung-homing domain or inhibiting Metadherin with siRNA has been reported to suppress breast cancer metastasis.
