Product Specific Information

The UCHT1 monoclonal antibody reacts with human CD3e, a 20 kDa subunit of the TCR complex. The epsilon chain, along with CD3 gamma and delta, is essential for proper assembly and surface expression of the TCR complex. CD3 is expressed by thymocytes and mature T cells.

Crosslinking of TCR via immobilized UCHT1 initiates intracellular signaling leading to activation and proliferation.

Each product contains:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-titrated for flow cytometry on human peripheral blood cells (5 µL/test).
A test corresponds to staining a cell sample in 100 µL volume, with 10⁵–10⁸ cells/test.

NovaFluor dyes are not compatible with DNA-intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability assays.

CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) is required for optimal staining with NovaFluor conjugates.
Add 5 µL per sample prior to combining with cells. For single-color controls, use 5 µL per 100 µL cell sample (10³–10⁸ cells).

NovaFluor conjugates utilize Phiton technology, based on nucleic acid dye structures engineered for minimal spectral spillover and spread.

Target Information

The CD3 complex transduces antigen-recognition signals into the cytoplasm of T cells and regulates TCR expression. It consists of CD3 gamma, delta, epsilon, and zeta subunits, encoded on chromosome 11. These subunits contain cytoplasmic tails with tyrosine-based motifs that mediate signal transduction.

CD3 is expressed on:

• 68–82% of peripheral blood lymphocytes
• 65–85% of thymocytes
• Purkinje cells in the cerebellum

It is not expressed on B or NK cells.
Defects in the CD3 gene can cause CD3 immunodeficiency.
