This ebic6 monoclonal antibody reacts with human Epstein-Barr Virus Induced Gene 3 (EBI3).
EBI3 can be expressed and secreted as monomers or homodimers and was first identified in virally infected B cells. It is also secreted by trophoblast cells, potentially contributing to immune tolerance during pregnancy.

EBI3 forms heterodimers with:

• p28 (IL-30) → forming IL-27, influencing Th1 differentiation and suppressing Th2/Th17 responses.
• p35 (IL-12 subunit) → forming IL-35, produced by regulatory T cells with immunosuppressive functions.

ELISA data from human dendritic cells suggest that EBI3 detected by flow cytometry is largely dimerized with p28 (IL-27). Testing with ebic6 for IL-35 is ongoing.

Applications Reported: Intracellular staining followed by flow cytometric analysis.
Applications Tested: Pre-titrated and validated using the Intracellular Fixation & Permeabilization Buffer Set (Product # 88-8824) and BestProtocols® (Protocol A).

Recommended Use: 5 µL (0.03 µg) per test (100 µL final volume; 10⁵–10⁸ cells/test).
Excitation: 488–561 nm; Emission: 578 nm; Laser: Blue, Green, Yellow-Green.
Filtration: 0.2 µm post-manufacturing filtered.

Target Information

EBI3 (Epstein-Barr Virus Induced-3) is a secreted glycoprotein that regulates Th2-type immune responses and tissue inflammation.
It dimerizes with IL-12 family subunits (p28 and p35) to form IL-27 and IL-35, respectively.
EBI3 expression is controlled by TLR signaling via MyD88 and NF-κB and plays roles in:

• Suppressing IL-17 and enhancing IL-10 production
• Regulating allergic airway inflammation
• Controlling tumor metastasis through CD8+ T cells
