Product Specific Information
The 17A2 monoclonal antibody reacts with the mouse CD3 complex. CD3 subunits gamma, delta, and epsilon are required for proper assembly, trafficking, and surface expression of the TCR complex. CD3 is expressed by thymocytes in a developmentally regulated manner and by all mature T cells. Binding of 17A2 to CD3 initiates intracellular signaling leading to activation and proliferation.
Applications Reported: Flow cytometric analysis
Applications Tested: Tested by flow cytometric analysis of mouse splenocytes. Recommended ≤1.0 µg/test (100 µL final volume).
Fluorochrome Information:
Brilliant Violet™ 650 (BV650) emits at 649 nm and is intended for use on cytometers equipped with a violet (405 nm) laser. Ensure instrument compatibility.
Staining Recommendations:
Use Super Bright Complete Staining Buffer (SB-4401-42) or Brilliant Stain Buffer™ (00-4409-75) when using multiple polymer dye-conjugated antibodies to minimize non-specific interactions.
Light Sensitivity:
Protect vial and stained samples from light.
Fixation:
Samples can be stored in IC Fixation Buffer (00-8222-49) or 1-step Fix/Lyse Solution (00-5333-54) for up to 3 days at 4°C in the dark. BV650 is not compatible with methanol-based fixation.

Target Information
The CD3 subunit complex is crucial for transducing antigen-recognition signals into the cytoplasm of T cells and regulating TCR surface expression. The CD3 subunits (gamma, delta, epsilon, zeta) are structurally related members of the immunoglobulin superfamily encoded on chromosome 11. CD3 is expressed on most T cells, thymocytes, and Purkinje cells, but not on B or NK cells. Defects in CD3 genes are associated with CD3 immunodeficiency.
