Product Specific Information

The SK7 monoclonal antibody reacts with human and chimpanzee CD3e, a 20 kDa subunit of the TCR complex. CD3 epsilon, along with gamma and delta subunits, is required for proper assembly, trafficking, and surface expression of the TCR complex. CD3 is expressed on thymocytes and all mature T cells. SK7 and UCHT1 clones recognize overlapping epitopes, while OKT3 and SK7 bind distinct epitopes.

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-diluted and tested by flow cytometry of normal human peripheral blood cells. Use 5 µL (0.25 µg) per test in 100 µL final volume. Suitable for 10⁵–10⁸ cells/test.

Fluorochrome Information:
Brilliant Violet™ 650 (BV650) emits at 649 nm and is excited by a 405 nm violet laser. Ensure instrument compatibility.

Staining Buffer Recommendation:
When using multiple polymer dye-conjugated antibodies, use Super Bright Complete Staining Buffer (Product # SB-4401-42) or Brilliant Stain Buffer™ (Product # 00-4409-75) to reduce non-specific interactions.

Light Sensitivity:
Protect from light to prevent photo-induced oxidation.

Fixation:
Samples may be stored in IC Fixation Buffer (Product # 00-8222-49) or 1-step Fix/Lyse Solution (Product # 00-5333-54) for up to 3 days at 4°C in the dark. BV650 is not compatible with methanol-based fixation.

Target Information

The CD3 subunit complex transduces antigen-recognition signals into T cells and regulates TCR surface expression. It includes CD3 gamma, delta, epsilon, and zeta subunits encoded on human chromosome 11. These subunits associate with cytoplasmic signaling molecules via tyrosine-based motifs. CD3 is expressed on most T lymphocytes and thymocytes but not on B or NK cells. Mutations in CD3 genes can lead to immunodeficiency.
