Product Specific Information

The FJK-16s antibody reacts with Foxp3 (FORKHEAD BOX P3, SCURFIN, JM2) from mouse, rat, dog, porcine, bovine, and cat. Cross-reactivity with other proteins has not been determined. Foxp3 is a 49–55 kDa transcriptional regulator associated with regulatory T cells (Treg). The antibody epitope maps to amino acids 75–125 of mouse Foxp3.

Applications include immunohistochemistry (frozen and paraffin-embedded tissues). Recommended usage:

• Frozen mouse spleen: ≤5 µg/mL
• Paraffin-embedded tissue: ≤10 µg/mL with antigen retrieval (high or low pH)

Not validated for flow cytometry. Optimal dilution should be determined by the user.

Filter Recommendation:
For eFluor® 570 conjugates, use filters capturing 570 nm emission (e.g., Excitation 545/25, 565LP, Emission 605/70). Alexa Fluor® 555 or TRITC filters are also suitable.

Target Information

FOXP3 (Forkhead box protein 3) is a transcriptional regulator essential for immune homeostasis, expressed in CD4⁺CD25⁺ regulatory T cells. It serves as a key marker for Treg cells. Mutations in FOXP3 cause the scurfy phenotype in mice and IPEX syndrome in humans. FOXP3 expression is also regulated by p53 in response to DNA damage in certain cancer cells.
