Target Information
In the intact cell, DNA closely associates with histones and other nuclear proteins to form chromatin. Chromatin remodeling is a critical component of transcriptional regulation, largely influenced by the acetylation and deacetylation of nucleosomal histones. Acetylation increases accessibility to transcription factors, while deacetylation is linked to transcriptional silencing.
Several mammalian nuclear histone acetylases have been identified, including GCN5, PCAF (p300/CBP-associated factor), p300/CBP, HAT1, and TFIID subunit TAF II p250. Mammalian HDAC8, isolated from human kidney, is a histone deacetylase with unique tissue distribution and nuclear localization. It plays roles in tissue development and cancer etiology.
