Product Specific Information
The 14D3 monoclonal antibody reacts with human CD152 (CTLA-4), a protein structurally similar to CD28. CTLA-4 is expressed on activated T cells (and sometimes B cells) and binds CD80 (B7-1) and CD86 (B7-2) with higher affinity than CD28.
CTLA-4 and CD28 deliver opposing signals: CD28 acts as a costimulator, while CTLA-4 inhibits IL-2 secretion and T cell proliferation, restricting activation. The cytoplasmic region of CTLA-4 contains ER retention motifs, leading to intracellular localization of newly synthesized CTLA-4 in response to TCR signaling.
This antibody also recognizes rhesus monkey CTLA-4 and exhibits inhibitory activity.
Applications Reported: Suitable for intracellular staining followed by flow cytometric analysis.
Applications Tested: Pre-titrated and tested using intracellular staining and flow cytometry of stimulated human peripheral blood cells with the Intracellular Fixation & Permeabilization Buffer Set (Product #88-8824-00).
Recommended usage ≤ 0.06 µg/test (100 µL final volume). Adjust cell number empirically (10⁵–10⁸ cells/test). Antibody titration is recommended for optimal performance.
Due to intracellular localization, full detection of CTLA-4 may require both surface and intracellular assessment.
Target Information
CTLA4 (Cytotoxic T-Lymphocyte Antigen 4) is a CD28-family receptor primarily expressed on CD4+ T cells. It binds CD80 and CD86 with higher affinity than CD28, delivering inhibitory signals that suppress T cell activation. Intracellular CTLA4 is also found in regulatory T cells and contributes to their immunosuppressive function.
