Product Specific Information

The UC10-4B9 monoclonal antibody reacts with mouse CD152, also known as cytotoxic T lymphocyte antigen-4 (CTLA-4). CTLA-4 is structurally similar to CD28 and is expressed on activated T cells at low levels. It binds B7 family members CD80 (B7-1) and CD86 (B7-2) with higher affinity than CD28.

CTLA-4 and CD28 deliver opposing signals to T cells: CD28 acts as a costimulator, while CTLA-4 inhibits IL-2 secretion and cellular proliferation, restricting T cell activation. The cytoplasmic portion of CTLA-4 contains ER retention motifs, leading to intracellular localization of newly synthesized CTLA-4 following TCR signaling.

Due to the intracellular localization of CTLA-4, complete detection may require assessment of both intracellular and surface expression.

Applications Reported: Flow cytometric analysis.
Applications Tested: Tested on resting and activated mouse splenocytes. Recommended use ≤1 µg/test (100 µL final volume). Cell numbers may range from 10⁵–10⁸ cells/test. Optimize titration for best performance.


Target Information

CTLA4 (Cytotoxic T-Lymphocyte Antigen 4) is a CD28-family receptor mainly expressed on CD4⁺ T cells. It binds CD80 and CD86 with higher affinity than CD28, but unlike CD28, CTLA4 inhibits T cell activation and function. Intracellular CTLA4 is also present in regulatory T cells and contributes to their function.
