Heat-mediated antigen retrieval is recommended prior to staining, using a 10 mM citrate buffer (pH 6.0) for 10–20 minutes, followed by cooling at room temperature for 20 minutes.
Following antigen retrieval, incubate samples with primary antibody for 30 minutes at room temperature.
A suggested positive control is liver or breast carcinoma.

Target Information

Folylpoly-gamma-glutamate synthetase (FPGS) catalyzes the addition of several equivalents of glutamic acid to the gamma-carboxyl group in the side chain of folate cofactors and analogs.
FPGS plays three key roles in folate homeostasis:

1. Polyglutamation prevents efflux of folate cofactors from the cell.
2. It increases the binding of folate cofactors to enzymes involved in folate interconversion and biosynthesis.
3. It enables accumulation of folates in mitochondria required for glycine synthesis.

FPGS is widely expressed in human tumors and is associated with cellular proliferation or lack of differentiation.
The cytotoxicity of thymidylate synthase and purine inhibitors requires continued inhibition of the target beyond one generation time, making FPGS integral to the efficiency of folate antimetabolites.
