Product Specific Information

The monoclonal antibody 20Raj1 recognizes the human Ki-67 protein, which exists in two isoforms (345 and 395 kDa) expressed in dividing cells. Ki-67 is detectable during active phases of the cell cycle (G1, S, G2, and mitosis) but absent in resting cells (G0).
During interphase, expression localizes to the nucleus and redistributes to chromosomes during mitosis, associating with heterochromatin-bound proteins such as CBX3.
In tumor studies, Ki-67 is used as a marker to determine the fraction of proliferating cells. This antibody also recognizes canine Ki-67.

Applications Reported: Intracellular staining followed by flow cytometric analysis.
Applications Tested: Pre-diluted and tested using human peripheral blood cells with the Foxp3/Transcription Factor Staining Buffer Set (Product #00-5523-00).
Recommended usage: 5 µL (0.25 µg) per test in a final volume of 100 µL. Cell number range: 10⁵–10⁸ cells/test.

Fluorochrome Information:
Brilliant Ultra Violet™ 395 (BUV395) emits at 395 nm and is intended for UV (355 nm) laser-equipped cytometers. Ensure instrument compatibility.

Buffer Recommendation:
When using multiple polymer dye-conjugated antibodies, use Super Bright Complete Staining Buffer (Product #SB-4401-42) or Brilliant Stain Buffer™ (Product #00-4409-75) to minimize non-specific interactions.

Target Information

Ki-67 is a nuclear protein expressed during late G1, S, G2, and M phases. It associates with euchromatin and heterochromatin through its FHA domain and shows cell cycle-dependent localization: perinucleolar in G1, nuclear matrix in G2, and chromosomal in M phase.
Used as a proliferation marker, Ki-67 is absent in G0 cells and increases from G1 through mitosis. It helps determine the growth fraction in neoplasms, correlating with tumor grade. Commonly used as a neuronal marker of cell cycling and proliferation.
