Target Information
Amyloid beta peptide (Abeta/Beta-amyloid)는 Alzheimer’s disease 환자의 뇌에서 발견되는 아밀로이드 플라크의 주요 구성 성분입니다. Abeta는 40–43개의 아미노산으로 구성되며, beta-amyloid precursor protein (beta APP)에서 두 단계의 절단 과정을 통해 생성됩니다. 첫 번째 단계는 beta-secretase (BACE)에 의해 beta APP의 아미노 말단 도메인이 절단되는 과정이며, 이후 gamma-secretase가 막 내 절단을 수행하여 amyloid beta peptide를 생성합니다. Gamma-secretase는 presenilin-1 및 2를 포함한 다단위 복합체로 구성되어 있으며, nicastrin과 결합하여 Abeta 생성 조절에 관여합니다. Abeta는 Alzheimer’s disease뿐 아니라 Lewy body dementia, Down syndrome, cerebral amyloid angiopathy 등에서도 검출됩니다.
