Target Information
Amyloid beta peptide (Abeta/Beta-amyloid)는 Alzheimer’s disease 환자의 뇌에서 발견되는 아밀로이드 플라크의 주요 구성 성분입니다. Abeta는 40–43개의 아미노산으로 이루어져 있으며, beta-amyloid precursor protein (beta APP)으로부터 두 단계의 절단 과정을 통해 생성됩니다.
- 첫 단계는 beta APP의 아미노 말단 영역을 beta-secretase (BACE)에 의해 절단하는 과정입니다. BACE는 propeptide 형태로 합성되어 furin에 의해 활성형으로 변환됩니다.
- 두 번째 단계는 gamma-secretase에 의해 막 내 절단이 일어나며, 이로써 amyloid beta peptide가 생성됩니다.
Gamma-secretase는 presenilin-1, presenilin-2, nicastrin 등을 포함하는 복합체로 알려져 있습니다. Amyloid beta는 Alzheimer’s disease, Lewy body dementia, Down’s syndrome, amyloidosis (Dutch type), cerebroarterial amyloidosis (cerebral amyloid angiopathy), Guam Parkinson-Dementia complex 등 다양한 신경퇴행성 질환에서 발견됩니다.
