• For immunohistochemistry (PFA fixed): incubate antibody for 30 minutes at room temperature.
• Staining of formalin-fixed tissues requires heating tissue sections in 10 mM Tris with 1 mM EDTA (pH 9.0) for 45 minutes at 95°C, followed by cooling at room temperature for 20 minutes.

Target Information

Amyloid beta peptide (Abeta/Beta-amyloid)는 알츠하이머병 환자의 뇌에서 발견되는 아밀로이드 플라크의 주요 구성 성분입니다.
Abeta 펩타이드는 40–43개의 아미노산으로 구성되며, β-amyloid precursor protein (βAPP)으로부터 β-secretase(BACE) 및 γ-secretase 효소에 의해 두 단계로 절단되어 생성됩니다.
이 과정에서 βAPP의 세포외 도메인이 제거되고, 막 결합 잔여 단백질이 γ-secretase에 의해 절단되어 amyloid beta peptide를 형성합니다.
γ-secretase 복합체에는 presenilin-1/2와 nicastrin이 포함되어 있으며, amyloid β 생성 조절에 관여합니다.
Amyloid β는 알츠하이머병뿐 아니라 Lewy body dementia, Down syndrome, cerebral amyloid angiopathy 등에서도 발견됩니다.
