Product Specific Information

The 1E7.2 antibody reacts with human and mouse ZAP-70, a cytosolic protein tyrosine kinase (PTK) belonging to the Syk family. ZAP-70 is expressed in T and NK cells and is essential for TCR signaling and development. It binds to phosphorylated ITAMs in the TCR complex and becomes phosphorylated by Lck and through autophosphorylation, enhancing its kinase activity and downstream signaling.

Mutations in ZAP-70 cause Severe Combined Immunodeficiency Syndrome (SCID) in humans. This antibody was generated against a KLH-peptide sequence corresponding to human ZAP-70 amino acid residues 282–307. ZAP-70 expression correlates with mutated IgVH expression in B-chronic lymphocytic leukemia (B-CLL), serving as a prognostic marker measurable by intracellular flow cytometry.

Applications Reported: Intracellular flow cytometric analysis.
Applications Tested: Tested on mouse thymocytes and human Jurkat cells using the Foxp3/Transcription Factor Buffer Set (Product #00-5523-00) and BestProtocols® Protocol B for intracellular proteins. Use ≤1 µg/test (100 µL final volume). Optimize titration per assay.

Target Information

ZAP-70 is a cytosolic protein tyrosine kinase of the Syk family, expressed in T-cells and NK cells. It is phosphorylated upon TCR engagement at tyrosines 292, 315, and 319, enhancing signaling via Vav and Lck binding sites, while phosphorylation at tyrosine 292 attenuates activation.
