Target Information
Glutathione peroxidases (GPXs) are ubiquitously expressed enzymes that catalyze the reduction of hydrogen peroxides and organic hydroperoxides by glutathione. Several isoforms exist with distinct structures and localizations:
- GPX1 (cGPx): Classical cytosolic/mitochondrial, selenium-dependent enzyme and the first discovered GPX family member.
- GPX2 (GI-GPx): Expressed in the gastrointestinal epithelium.
- GPX3 (pGPx): Extracellular plasma form, mainly expressed by the kidney and released into circulation.
- GPX4 (PH-GPx): Reduces a variety of hydroperoxides, including membrane-integrated hydroperoxides and lipid peroxides in LDL.
Most mammalian GPX members, except for Cys-containing GPX3 and epididymis-specific GPX5, possess selenocysteine at the active site.
