Target Information
Peroxiredoxin (Prx) is a peroxidase family involved in cell proliferation, differentiation, and apoptosis. Mammalian Prx proteins (approximately 50 isoforms) are classified by sequence homology and cysteine-based active sites.
Mammalian Prxs include 6 members grouped into typical 2-Cys, atypical 2-Cys, and 1-Cys Prx. Except for Prx VI (1-Cys subgroup), the other five 2-Cys isotypes exhibit thioredoxin-dependent peroxidase (TPx) activity using thioredoxin, thioredoxin reductase, and NADPH as reducing systems.
Mammalian Prxs range from 20–30 kDa and vary in subcellular localization:
- Prx I, II, VI: cytosol
- Prx III: mitochondria
- Prx IV: ER and secretion
- Prx V: peroxisome, mitochondria, and cytosol
