Target Information
Estrogen Receptors (ER) are members of the steroid/thyroid hormone receptor superfamily of nuclear receptors. The estrogen receptor is a ligand-activated transcription factor that, when bound to estrogen, undergoes a conformational change enabling dimerization and binding to estrogen response elements (ERE) in DNA. This interaction regulates gene transcription through coactivator or corepressor recruitment.
There are two forms of the estrogen receptor, alpha (ESR1) and beta (ESR2), encoded by separate genes. At least four isoforms of ER-alpha are known (66 kDa, 53 kDa, 47 kDa, 35 kDa). Estrogen receptors are widely expressed across tissues and are critical for sexual development, reproductive function, and bone physiology. They are also implicated in pathological processes such as breast cancer, endometrial cancer, and osteoporosis.
