Target Information
Estrogen Receptors (ER) are members of the steroid/thyroid hormone receptor superfamily of nuclear receptors. The estrogen receptor is a ligand-activated transcription factor that, when bound to estrogen, undergoes a conformational change enabling dimerization and binding to estrogen response elements (ERE) in DNA. This interaction allows ER to regulate gene transcription positively or negatively by recruiting coactivator or corepressor proteins.
Two distinct forms of estrogen receptor exist — alpha (ESR1) and beta (ESR2) — encoded by separate genes. Alternative RNA splicing produces multiple isoforms of estrogen receptor alpha (66 kDa, 53 kDa, 47 kDa, 35 kDa). Estrogen receptors are widely expressed in various tissues, essential for sexual development and reproductive function, and play roles in bone and other tissues. Dysregulation is associated with diseases such as breast cancer, endometrial cancer, and osteoporosis.
