Product Specific Information

The 8.1.1 monoclonal antibody reacts with mouse podoplanin (T1a, gp38, aggrus), a 43 kDa transmembrane glycoprotein expressed in kidney glomerular epithelial cells (podocytes). Podoplanin is also expressed in epithelial and mesothelial cells such as intestinal epithelium, alveolar type I cells, podocytes, and visceral peritoneum mesothelium. It serves as a potent marker for lymphatic endothelium and is expressed by subcapsular epithelial cells of the murine thymus. Mice deficient in Podoplanin die at birth due to respiratory defects and congenital lymphedema resulting from lymphatic pattern formation failure.

Applications Reported: Flow cytometric analysis
Applications Tested: Tested on TE-71 cell line at ≤0.25 µg/test (100 µL final volume; 10⁵–10⁸ cells/test).
Recommended Laser: Blue, Green, or Yellow-Green Laser
Excitation/Emission: 488–561 nm / 578 nm
Filtration: 0.2 µm post-manufacturing filtered

Target Information

Podoplanin (PDPN, T1A, gp38, Aggrus) is a mucin-type glycoprotein (40–43 kDa) expressed in various tumors and normal tissues, especially lymphatic endothelial cells and follicular dendritic cells. It localizes in stromal cells of peripheral lymphoid tissue and thymic epithelial cells. As a regulator of lymphatic endothelium, podoplanin helps maintain podocyte morphology and serves as a ligand for CLEC-2. Expression correlates with tumor invasiveness and metastatic potential. It is involved in cell migration, metastasis formation, and tumor invasion. Podoplanin expression is induced via Src-Cas signaling and is also found on Th17 cells. Dysfunction is linked to diseases such as lymphangioma and subglottis neoplasm and has been proposed as a marker for lung injury.
