Product Specific Information

Specificity: This antibody binds to mouse PD-L2 and competes with PD-1 binding. It does not cross-react with PD-L1.

Target Information

Programmed death-ligand 2 (PD-L2, B7-DC) is a member of the B7 ligand family within the immunoglobulin superfamily and acts as a ligand for PD-1. PD-L2 is primarily expressed in dendritic cells but can be induced in various immune and non-immune cells depending on the microenvironment. Expression is upregulated by Th2 cytokine IL-4 and to a lesser extent by Th1 cytokines TNF-α and IFN-γ.

PD-L2 generally shows lower expression compared to PD-L1 but has 2–6 times higher affinity for PD-1. The PD-1/PD-L pathway functions as an inhibitory immune checkpoint, regulating T cell activation, proliferation, and cytokine production (e.g., IFN-γ, IL-10). Interestingly, PD-L2 can also stimulate T cell responses independently of PD-1, indicating additional receptor interactions.

Recent studies identified repulsive guidance molecule b (RGMb) as a second receptor for PD-L2. RGMb, expressed in the CNS and macrophages, plays a role in respiratory tolerance through BMP and/or neogenin signaling pathways. The human PD-L2 monomer consists of a 201-amino-acid extracellular domain, a 21-amino-acid transmembrane domain, and a 32-amino-acid cytoplasmic domain.
