Product Specific Information

This NGZB monoclonal antibody reacts with mouse Granzyme B, a member of the granzyme serine protease family found in cytotoxic T cells and NK cells. Granzyme B, also known as CGL1 or CTLA-1, is expressed only in activated cytotoxic T lymphocytes and plays a key role in apoptosis of target cells via mannose 6-phosphate receptor-mediated pathways.

  • Applications Reported: Suitable for intracellular staining and flow cytometric analysis.
  • Applications Tested: Validated using stimulated mouse splenocytes and the Intracellular Fixation & Permeabilization Buffer Set (Product # 88-8824-00).
  • Recommended usage ≤0.125 µg/test (100 µL final volume).
  • eFluor® 660 is a replacement for Alexa Fluor® 647 and emits at 659 nm when excited with a 633 nm red laser.
  • Filtration: 0.2 µm post-manufacturing filtered.

Target Information

Granzyme B is a serine protease located in cytotoxic T cells and NK cell granules. It induces apoptosis in target cells through caspase-3 activation and mitochondrial pathways (via Bid cleavage). It interacts with mannose 6-phosphate receptor as a death receptor during cytotoxic T cell-mediated apoptosis. Multiple granzymes (A–G) have been identified in mouse CTLs and NK cells, while fewer have been cloned in humans.
